5-Amino-1MQ

5-Amino-1MQ (5-Amino-1-methylquinolinium) is a selective small-molecule inhibitor of NNMT (Nicotinamide N-methyltransferase), an enzyme highly expressed in adipose tissue that catalyzes the methylation of nicotinamide using S-adenosylmethionine (SAM) as the methyl donor, producing 1-methylnicotinamide and S-adenosylhomocysteine (SAH) as products.

By inhibiting NNMT, 5-Amino-1MQ conserves both NAD+ precursor availability (preventing nicotinamide consumption) and SAM (preventing methylation capacity depletion), effectively reversing the epigenetic and metabolic reprogramming that drives adipogenesis and obesity. Preclinical studies have demonstrated significant reductions in fat mass, adipocyte size, and weight gain without caloric restriction, making it a novel research tool for studying NNMT-mediated epigenetic control of metabolic disease.
