Overview
Adipotide (CKGGRAKDC-GG-D(KLAKLAK)2) is a chimeric peptidomimetic consisting of two functional domains connected by a GG linker: a targeting domain (CKGGRAKDC) that binds prohibitin on the vascular endothelium of white adipose tissue blood vessels, and a pro-apoptotic effector domain (D(KLAKLAK)2) — an amphipathic D-amino acid sequence that disrupts mitochondrial membranes upon cell internalization.
The targeting domain exploits the differential expression of prohibitin on adipose vasculature versus other tissues, enabling relatively selective delivery of the mitochondria-disrupting effector domain to fat-feeding blood vessels. Apoptosis of adipose vasculature leads to reduced blood supply and secondary adipocyte death. In published non-human primate studies, adipotide produced significant decreases in body weight (11%) and abdominal fat (27%) without general systemic toxicity, representing a research model for targeted white adipose tissue ablation.
