Overview
NAD+ (Nicotinamide Adenine Dinucleotide) is a fundamental coenzyme present in all living cells, serving two critical functions: as a redox carrier in cellular energy metabolism (accepting and donating electrons in oxidative phosphorylation), and as a substrate for a growing class of enzymes including sirtuins (SIRT1–7), poly(ADP-ribose) polymerases (PARPs), and CD38/CD157 ectoenzymes.
NAD+ levels decline significantly with age (by up to 50% between young adulthood and middle age) and are associated with impaired mitochondrial function, reduced DNA repair capacity, and metabolic dysfunction. Research into NAD+ supplementation focuses on its potential to activate sirtuins (particularly SIRT1 and SIRT3), promote mitochondrial biogenesis, enhance DNA repair, modulate circadian rhythms, and extend healthspan in model organisms.
