ActRIIB decoy — traps myostatin to remove the brake on muscle growth.
Overview
ACE-031 is a soluble decoy receptor fusion protein constructed from the extracellular ligand-binding domain of Activin Receptor Type IIB (ActRIIB) fused to the Fc region of human IgG1. The ActRIIB ectodomain has nanomolar affinity for multiple TGF-β superfamily ligands that negatively regulate muscle mass, most notably myostatin (GDF-8), activin A, GDF-11, and BMP-9.
By sequestering these ligands before they can engage cell-surface ActRIIB receptors and activate the SMAD2/3 signaling pathway, ACE-031 effectively removes the brakes on muscle protein synthesis, resulting in significant increases in lean muscle mass. In Phase I/II clinical trials in healthy volunteers, ACE-031 produced dose-dependent increases in thigh muscle volume within weeks of administration. It has been studied in Duchenne muscular dystrophy, cancer cachexia, and other muscle-wasting conditions.
