Overview
ARA-290 is an 11-amino acid helix B surface peptide (HBSP) derived from the non-erythropoietic portion of erythropoietin (EPO). It is designed to selectively activate the Innate Repair Receptor (IRR) — a tissue-protective heteroreceptor complex consisting of the EPO receptor subunit (EPOR) and the common beta chain (CD131/βc) — without activating the homodimeric EPOR (EPOR2) complex responsible for erythropoiesis.
This receptor selectivity allows ARA-290 to deliver the cytoprotective, anti-inflammatory, and neuroregenerative benefits of EPO without the hematopoietic side effects (thrombosis risk, polycythemia) that limit therapeutic EPO use. ARA-290 has undergone clinical trials for painful small fiber neuropathy (SFN) in sarcoidosis patients, where it improved corneal nerve fiber density and pain scores, providing human clinical validation for its neuroprotective mechanism.
