FOX04-DRI

FOXO4-DRI is a retro-inverso D-amino acid peptide designed to disrupt the interaction between the FOXO4 transcription factor and p53 specifically in senescent cells. In normal physiology, senescent cells upregulate FOXO4, which sequesters p53 in the nucleus and prevents it from triggering apoptosis — effectively allowing senescent cells to persist and contribute to the senescence-associated secretory phenotype (SASP).

The FOXO4-DRI peptide competitively disrupts this FOXO4–p53 interaction, freeing p53 to translocate to mitochondria and initiate apoptosis selectively in senescent cells. Critically, healthy proliferating cells, which have low FOXO4 expression, are largely spared. This selectivity makes FOXO4-DRI one of the most mechanistically specific senolytic compounds studied to date, with published research demonstrating improved physical function and fur coat restoration in aging mice.
