Overview

KPV (Lys-Pro-Val) is the C-terminal tripeptide of alpha-melanocyte-stimulating hormone (α-MSH) that retains the cytokine-inhibiting anti-inflammatory activity of the parent hormone while being devoid of its melanotropic effects. Unlike full-length α-MSH, KPV does not activate cell-surface melanocortin receptors — instead, it enters cells through a specific receptor-independent uptake mechanism and acts intracellularly.

Once internalized, KPV inhibits NF-κB activation, downregulating the transcription of pro-inflammatory genes including TNF-α, IL-1β, IL-6, and IL-8. This intracellular anti-inflammatory mechanism is distinct from receptor-mediated approaches and has been studied particularly in intestinal inflammation models (IBD, colitis, gut barrier dysfunction) and skin inflammation, where KPV demonstrates efficacy via topical and oral routes in addition to parenteral administration.
