Product Overview

PE-22-28 is a synthetic truncated analog of Spadin, the active peptide fragment (residues 22–28) derived from the propeptide of human sortilin (a vacuolar protein sorting receptor). Spadin was identified as an endogenous antagonist of TREK-1 (TWIK-related K+ channel 1, encoded by KCNK2), a two-pore domain background potassium channel with high expression in hippocampal and cortical neurons.

TREK-1 is a mechanosensitive, temperature-sensitive, and lipid-regulated channel that normally acts to stabilize neuronal resting membrane potential. Its blockade by PE-22-28 produces rapid (24–48 hour onset) antidepressant-like effects in preclinical models — a pharmacological profile distinct from classical antidepressants that require weeks of administration. Research also indicates TRPV4 modulatory activity, adding to its mechanistic complexity.

Research Applications

PE-22-28 has been examined in experimental studies related to:

  • TREK-1 (KCNK2) potassium channel antagonism and neurophysiology
  • Rapid-onset antidepressant mechanism research (24–48h)
  • Spadin peptide analog pharmacology and structure-activity studies
  • TRPV4 modulation and mechanosensitive channel research
  • Depression and mood disorder preclinical models
  • Comparison with classical antidepressants on onset kinetics

Handling and Storage (Laboratory Guidance)

This product is supplied as a lyophilized peptide powder.

Typical laboratory handling recommendations include:

  • Store at 2–8 °C upon receipt.
  • Protect from heat, moisture, and direct light.
  • Avoid excessive agitation of the solution to maintain peptide stability.
  • Reconstitution, if required for laboratory experiments, should be performed using appropriate sterile laboratory techniques.
  • Use of acetic acid water (0.6%) or sterile bacteriostatic water is recommended for peptides with low aqueous solubility.
