Overview
PE-22-28 is a synthetic truncated analog of Spadin, the active peptide fragment (residues 22–28) derived from the propeptide of human sortilin (a vacuolar protein sorting receptor). Spadin was identified as an endogenous antagonist of TREK-1 (TWIK-related K+ channel 1, encoded by KCNK2), a two-pore domain background potassium channel with high expression in hippocampal and cortical neurons.
TREK-1 is a mechanosensitive, temperature-sensitive, and lipid-regulated channel that normally acts to stabilize neuronal resting membrane potential. Its blockade by PE-22-28 produces rapid (24–48 hour onset) antidepressant-like effects in preclinical models — a pharmacological profile distinct from classical antidepressants that require weeks of administration. Research also indicates TRPV4 modulatory activity, adding to its mechanistic complexity.
