Overview
SLU-PP-332 (a.k.a. SLU-PP-332) is a potent synthetic pan-agonist of Estrogen-Related Receptors alpha, beta, and gamma (ERRα/β/γ) — orphan nuclear receptors in the nuclear receptor superfamily that lack known endogenous ligands but play critical roles in regulating oxidative metabolism, mitochondrial biogenesis, and energy homeostasis in high-energy tissues including heart, skeletal muscle, and brown adipose tissue.
SLU-PP-332 binds to and activates all three ERR isoforms simultaneously, triggering transcriptional programs that upregulate PGC-1α-dependent mitochondrial biogenesis, oxidative phosphorylation, and fatty acid oxidation pathways. In preclinical studies, SLU-PP-332 produced remarkable exercise-mimetic effects including significant increases in running endurance (70% improvement in mice) and demonstrated therapeutic potential in heart failure models by restoring cardiac metabolic function and reducing pathological cardiac remodeling.
