Overview
IGF-1 LR3 (Insulin-like Growth Factor-1 Long Arg3) is a modified analog of endogenous IGF-1 engineered with two key changes: an arginine-3 substitution (replacing the native glutamate) and a 13 amino acid N-terminal extension. These modifications dramatically reduce binding affinity to IGF-binding proteins (IGFBPs 1–6), which normally act as transport and regulatory proteins for IGF-1 in circulation.
By reducing IGFBP binding, IGF-1 LR3 remains in its free, biologically active form significantly longer than native IGF-1, with an estimated half-life of 20–30 hours compared to ~12 minutes for native IGF-1. This extended activity makes it a powerful tool for studying the IGF-1 receptor (IGF-1R) signaling axis, cellular proliferation, differentiation, and anabolic metabolism in research settings.
