Overview

VIP (Vasoactive Intestinal Peptide) is a 28-amino acid neuropeptide belonging to the glucagon/secretin superfamily, expressed in neurons of both the central and peripheral nervous systems as well as in immune cells and intestinal mucosa. It acts on two G-protein coupled receptors, VPAC1 (high expression in brain, intestine, and immune cells) and VPAC2 (expression in smooth muscle and brain), both signaling through cAMP/PKA pathways.

VIP exerts a wide range of regulatory effects: it is a potent vasodilator and bronchodilator in the pulmonary and gastrointestinal systems, modulates circadian rhythm timing through the suprachiasmatic nucleus (SCN), and demonstrates significant anti-inflammatory activity by suppressing TNF-α, IL-6, and IL-12 production while stimulating IL-10. It is studied in models of autoimmunity, neuroinflammation, pulmonary disease, and gastrointestinal physiology.
