Overview
MOTS-c (Mitochondrial Open reading frame of the Twelve S rRNA type-c) is a 16-amino acid mitochondria-derived peptide (MDP) encoded within the 12S ribosomal RNA gene of the mitochondrial genome — making it one of a rare class of peptides encoded by mitochondrial rather than nuclear DNA.
MOTS-c acts as a metabolic regulator through AMPK (AMP-activated protein kinase) activation, enhancing glucose uptake in skeletal muscle, improving insulin sensitivity, and promoting mitochondrial homeostasis. Remarkably, plasma MOTS-c levels have been found to be significantly higher in long-lived individuals in human studies, and exercise increases MOTS-c production, suggesting it may function as a mitokine mediating exercise-related metabolic benefits.
