Overview
Melatonin (N-acetyl-5-methoxytryptamine) is an indoleamine neurohormone synthesized from serotonin (via AANAT and HIOMT enzymes) primarily in the pineal gland, with production tightly controlled by light exposure through the suprachiasmatic nucleus (SCN). Its rhythmic nocturnal secretion is the primary biochemical signal communicating darkness and time-of-day information to the body.
Melatonin acts on MT1 and MT2 G-protein coupled receptors in the SCN, pituitary, and peripheral tissues to synchronize circadian rhythms, promote sleep onset, and regulate seasonal biology. Beyond chronobiology, melatonin is one of the most potent endogenous antioxidants known — directly scavenging free radicals and upregulating antioxidant enzymes (SOD, GPx, catalase). Research also examines its roles in mitochondrial protection, immune modulation, and anti-tumor biology.
